A series of AcNPV mutants with deletion located at upstream region of the polyhedrine promoter were generated and the effects of deletions on the promoter strength were examined by analyzing the activity of the transiently expressed CAT gene

to the polyhedrin promoter, Deletion from BamHI to EcoRV(from +35 to -95) region containing TATA box, eight base pairs encompassing the transcription initiation site, and 5' non-coding sequences abolished the promoter activity but deletions from
EcoRV
to Sall(from -95 to -1135), from SalI to SalI (from -825 to -1135), from EcoRV to XhoI(from -95 to -2105), and from HindIII to HIndIII(+2006 to +3086)did not reduce the prowoter stength sighiticautly. These resutts indicated that cis-acting
enhancing
element required for polyhedrin promoter strength is not
present within the deleted regions and confirm the previous reports that the essential sequences responsible for the polyhedrin promoter strength are located between transcription initiation site and translation initiation codon.
